This kit applies the principle of real-time PCR-fluorescence probing technology which design specific primer probes with two specific conserved regions of Mpox Virus, MPXV-1 and MPXV-2 gene. The 5' of MPXV-1 gene probe is labeled as FAM, while the 3' quencher group is BHQ1 and the 5' of MPXV-2 gene probe is labeled as VIC, while the 3' quencher group is BHQ1. During PCR amplification, specific primers and probes bind to their respective target sequences. The 5' exonuclease activity of Taq enzyme cleaves and degrades the probes, so that the reporter and the quencher fluorophore groups are separated, the fluorescence monitoring system will receive the fluorescent signal accordingly. Which is, each time a DNA strand is amplified, a fluorescent molecule is formed, resulting in the complete synchronization of the accumulation of fluorescent signals with the formation of PCR products. At the same time, endogenous internal reference quality control was introduced to avoid false negatives in the test.
This PCR Reagent product is a nucleic acid amplification reaction solution that includes a PCR reaction mixture and Polymerase Chain Reaction Reagent. It is designed to provide standard reaction conditions for the amplification of nucleic acid from a skin (mucosal) lesion site specimen. The MPXV Reagent Strip contains PCR Buffer, primers, Probes, DNTPs, Taq DNA Polymerase, and Uracil-N-Glycosylase. The Positive Control is Plasmid DNA.

The Real-Time PCR Kit for Detection of Monkeypox Virus is a qualitative in vitro real-time PCR test for the detection of nucleic acid from monkeypox virus in oropharyngeal swab, nasopharyngeal swab, human pustular or vesicular rash specimens from individuals suspected of monkeypox virus infectious.
Results are for the identification of monkeypox virus DNA. Positive results are indicative of the presence of monkeypox virus DNA; clinical correlation with patient history and other diagnostic information is necessary to determine patient infection status. Positive results do not rule out bacterial infection or co-infection with other viruses.
The agent detected may not be the definite cause of disease.
Negative results do not preclude monkeypox virus infection and should not be used as the sole basis for patient management decisions. Negative results must be combined with clinical observations, patient history, and epidemiological information.
The Real-Time PCR Kit for Detection of Monkeypox Virus is intended for use by qualified and trained clinical laboratory personnel specifically instructed and trained in the techniques of real-time PCR and in vitro diagnostic procedures.
