INTENDED USE
The kit is used for the quantitative detection of Human
Immunodeficiency Virus Type 1 (HIV-1) RNA in human serum or
plasma by Real time RT-PCR method. It can be used to evaluate
antiviral treatment and monitor the therapeutic effect by monitoring
HIV RNA baseline levels and changes in patient blood. It is not
intended for screening of blood and blood products for the presence
of HIV-1 RNA or confirmation of the diagnosis of infection with
HIV-1.
SUMMARY
HIV-1
is the etiologic agent of Acquired Immunodeficiency
Syndrome (AIDS). It can be transmitted through contact,
exposure to infected blood or blood products, prenatal infection of a
fetus, or perinatal or postnatal infection of a newborn.
During the acute HIV infection stage, the level of HIV in the blood is
very high, which greatly increases the risk of HIV transmission.
During the chronic infection stage, HIV levels are very low in the
blood. People with chronic HIV infection may not have any
HIV-related symptoms. Without antiretroviral therapy, chronic HIV
infection usually advances to AIDS in 10 years or longer.
At AIDS stage, viral load and risk of transmission is very high.
Without treatment, people with AIDS typically survive about 3 years.
Quantitative measurements of HIV in the peripheral blood have
shown that higher virus levels may be correlated with increased risk
of clinical progression of HIV-associated disease, and shown that
reductions in plasma virus levels may be associated with decreased
risk of clinical progression.
PRINCIPLE
The gag gene region of HIV-1 genome is used to generate probes
and primers in this real-time RT-PCR assay. HIV-1 gag RNA are
reverse-transcribed and amplified accurately by the specific primers.
At the same time, the highly specific TaqMan probe can be combined
with the target nucleic acid fragments, and will be hydrolyzed to
produce the fluorescence signals under the action of Taq enzyme
exonuclease activity, in which the real-time amplification curve will be
obtained according to the relationship between the fluorescence
signal and the number of amplification cycles, besides, the false
negative and the PCR inhibition can be monitored by Internal Control.
The gag RNA (FAM channel) of HIV be quantitatively detected under
the quality control by internal control (ROX channel).
