APPLICATIONS:

The Diagnostic Kit for Human Parvovirus B19 IgM Antibody is a specialized in vitro diagnostic test designed to detect the presence of IgM antibodies specific to Parvovirus B19 in human serum or plasma. IgM antibodies are produced by the immune system as an early response to infection, indicating a recent or acute Parvovirus B19 infection.   1. Purpose:

    • Detects IgM antibodies against Parvovirus B19 in human serum or plasma samples.

A secondary antibody conjugated with an enzyme or chemiluminescent label is added to produce a measurable signal (color change or luminescence), indicating the presence of IgM antibodies.
  3. Sample Type:

    • Requires serum or plasma samples, typically collected through venous blood draw.
    • The sample is processed to separate the serum or plasma for testing.
  4. Assay Sensitivity and Specificity:

    • The kit is designed to have high sensitivity and specificity for IgM antibodies to Parvovirus B19.
    • Cross-reactivity with other viral infections is minimal, ensuring accurate results.
  5. Test Procedure:

    • The patient’s serum or plasma sample is added to a microplate well pre-coated with Parvovirus B19 antigens.
    • After incubation, unbound substances are washed away.
    • A detection antibody (often conjugated with an enzyme or luminescent marker) is added to bind to any Parvovirus B19 IgM antibodies present in the sample.
    • A substrate is then added, which reacts with the enzyme to produce a colorimetric or luminescent signal.
    • The intensity of the signal is proportional to the amount of IgM antibodies in the sample.
    • Results are compared to a standard curve or reference value to determine positivity.

PACKAGING:

Elisa test kits are carefully packaged and shipped in an insulated box to ensure the temperature is maintained during transport. The package contains the test kit, instructions, and other supplies needed for the test.

 

It also includes a return label for customers to return unused portions of the kit after testing is complete. All packages come with tracking information, so customers can track the progress of their shipment.
 
STORAGE OF THE TEST KIT:

Unopened test kits should be stored at 2-8C upon receipt and the microtiter plate should be kept in a sealed bag to minimize exposure to damp air. Use up the reagents as soon as possible after the kit is unpacked.
 
ASSAY PROCEDURE

  • Bring ELISA Kit (all reagents), and samples to room temperature before use (approximately 30 minutes).
  • Dilute concentrated wash buffer 1:19 with ddH2O.
  • For each test, set one blank, two positive and three negative controls. Add 100μl positive and negative control serum into positive and negative control wells respectively.
  • Add 100μl sample diluent into test wells, then add 10μl specimen in each well.
  • Cover wells with seal paper, then incubate 30 minutes at 37°C.
  • Wash each well 5 times by filling each well with diluted wash buffer, then inverting the plate vigorously to get all water out and blocking the rim of wells on absorbent paper for 15 seconds.
  • Add 100 μl Enzyme Conjugant to each well except the blank well.
  • Incubate: Cover the Microplate with plate cover and incubate the Coated Microplate in a thermostat-controlled water-bath or microplate incubator at 37℃ for 30 minutes.
  • Wash the Plate as step 6.
  • Add Substrate: Add 50μL of Substrate Solution A and 50μL of Substrate Solution B to each well, include blank. mix well. Cover and incubate at 37℃ for 15 minutes.
  • Stop reaction: Add 50μL Stop Solution to each well, include blank mix well.
  • Read the absorbance at 450 nm. If a dual wavelength measurement is used, the reference wavelength should be selected from 630nm to 690nm.

INTERPRETATION OF RESULTS:

  • Cut off Value calculation:
  • COV = 0.10 + the average OD of negative controls (If the absorbance of negative controls is below 0.05, calculate it as 0.05.If the absorbance of negative controls is above 0.05; calculate it as its original value.)
  • Positive OD450 of sample ≥ COV HPVB19- IgM
  • Negative OD450 of sample < COV. HPVB19- IgM
