The thyroid hormone thyroxine (T4) is physiologically part of the thyroid regulatory circuit and has an impact on systemic metabolism. The main part of total thyroxine is bound to transporters (TBG, prealbumin and albumin). Free thyroxine (fT4) is the physiologically active thyroxine component.
The determination of free thyroxine is an important part of routine clinical diagnosis. When thyroid dysfunction is suspected, free T4 and TSH are measured. The determination of fT4 is also suitable for monitoring thyroid suppressive therapy.

Test principle

Competition principle. Total test time: 80 minutes

• Sample, T4 derivative coated microwells and enzyme-labeled anti-T4 are combined.

• During the incubation period, the T4 derivative coated on the microwells and the FT4 present in the sample compete for binding to the enzyme-labeled antibody.

• After washing, a complex is generated between the solid phase and the enzyme-linked antibody by an immune reaction.

• The substrate solution is then added and catalyzed by this complex, resulting in a color reaction. The resulting color reaction is measured as absorbance.
