This kit is used to qualitatively detect IgM antibodies to Coxsackievirus A16 in human whole blood, serum or plasma samples. It is used as an auxiliary diagnostic reagent for Coxsackievirus A16 infection in clinical diagnosis. Coxsackievirus (CV) belongs to the genus Enterovirus in the family Picornaviridae. It is the causative agent of many human diseases with a wide spectrum of pathogenicity, including respiratory tract infection, conjunctivitis, febrile rash, hand, foot and mouth disease, herpetic pharyngitis, myocarditis, cardiomyopathy, aseptic meningitis, encephalitis and acute flaccid paralysis [2]. There are 30 serotypes of Coxsackievirus, which are divided into two groups, A and B. Among them, CV Group A 16 Strain (CVA16) is one of the main pathogens causing hand, foot and mouth disease in children. CVA16 has spread throughout the world and is mainly transmitted through the fecal, oral, respiratory and close contact routes. The main targets of infection are children ≤5 years old. It often co-circulates with enterovirus 71 (EV71)[2].
This kit uses the principle of colloidal gold immunochromatography technology. Colloidal gold-labeled mouse anti-human IgM is coated on the gold label pad. It is used to qualitatively detect Coxsackie virus A16 IgM antibodies in whole blood, serum or plasma samples. When testing positive samples, Coxsackie virus A16 IgM antibodies in the sample can bind to colloidal gold-labeled mouse anti-human IgM to form immune complexes. Due to the chromatography, the complex and the sample flow forward inside the nitrocellulose membrane. When the complex passes through the T line, it binds to the coated recombinant CVA16-Ag to form "Au-mouse anti-human IgM-CVA16-IgM-CVA16-Ag" and agglutinates and develops color. The remaining colloidal gold-labeled mouse anti-human IgM binds to the sheep anti-mouse IgG antibody coated at the C line to agglutinate and develop color. When testing negative samples, the sample does not contain Coxsackie virus A16 IgM antibodies, resulting in the inability to form immune complexes, and can only develop color at the C line.

1. Preparation before the test: 1 each of 10µl, 50µl and 100µl micropipettes and several matching tips or droppers (10µl, 50µl, 100µl).
2. Test process: Place the test card on a dry horizontal work surface. Use a micropipette to add 10µl of serum or plasma sample (50µl for whole blood) to each well of the test card, then add 100µl of sample diluent, and observe the results within 15-20 minutes.
3. Precautions during the test process: a. This kit and the sample to be tested should be placed at room temperature for equilibrium before testing. b. Use the test card within 30 minutes after opening c. The results observed after 20 minutes are invalid.
