LH (luteinizing hormone) belongs to the gonadotropin family along with FSH (follicle-stimulating hormone). LH and FSH work together to regulate and stimulate the growth and function of the gonads (ovaries and testes). Like FSH, TSH, and hCG, LH is a glycoprotein composed of two subunits (alpha and beta chains). (1, 2, 3) This protein hormone consists of 121 amino acids and three sugar chains with a molecular weight of 29,500 daltons. In women, gonadotropins act in the hypothalamic-pituitary-ovarian regulatory circuit to control the menstrual cycle. (4, 5) LH and FSH are released in pulses from the gonadotropins of the anterior pituitary and transported to the ovaries via the bloodstream. Here, gonadotropins stimulate the growth and maturation of ovarian follicles, thereby promoting the biosynthesis of progesterone. The highest LH concentration occurs in the middle of the menstrual cycle and induces ovulation and the formation of the corpus luteum, the main secretory product of which is progesterone. In the Leydig cells of the testes, LH stimulates the production. Measurement of LH concentrations can be used to elucidate dysfunction within the hypothalamic-pituitary-gonadal system. Combined measurement of LH and FSH is useful in the following indications: congenital disorders with chromosomal aberrations (e.g., Turner syndrome), polycystic ovaries (PCO), established amenorrhea, menopausal syndrome, and suspected causes of Leydig cell insufficiency
Sandwich principle. Total test time: 80 minutes.
• The sample, anti-LH coated microwells and enzyme-labeled anti-LH are mixed together.
• During the incubation, LH in the sample can react with both antibodies simultaneously, resulting in the LH molecule being sandwiched between the solid phase and the enzyme-linked antibody.
• After washing, a complex is formed between the solid phase, LH in the sample and the enzyme-linked antibody by an immune reaction.
• Then a substrate solution is added and catalyzed by this complex, resulting in a color reaction. The resulting color reaction is measured as absorbance.
• The absorbance is proportional to the amount of LH in the sample.

• All products containing human serum or plasma were found to be non-reactive to HBsAg, HCV, and HIVI/II. However, all products should be considered as potential biohazards during use and disposal.
• Mix the samples in the wells thoroughly by shaking and eliminate air bubbles.
• Perform the assay away from harsh environmental conditions. For example, ambient air containing high concentrations of corrosive gases (such as sodium hypochlorite, alkaline, acetaldehyde, etc.) or dusty air.
• Wash the wells thoroughly. Each well must be completely filled with the wash solution.
However, the injection force should not be too strong to avoid overflow. During each wash cycle, wipe the liquid in each well dry. Tap the microplate on absorbent paper to remove residual water droplets. It is recommended to use an automated microplate strip washer to wash the microplate.
• Failure to adequately remove adhering solutions during the aspiration or decantation wash steps may result in poor replication and inaccurate results.
• Do not use valid reagents that have expired after the labeled expiration date.
• Do not mix or use components from kits with different batch codes.
• If using multiple plates, it is recommended to repeat the calibration curve.
• To obtain reproducible results, the reaction time must remain constant in each well.
• Ensure that the bottom of the plate is clean and dry.
• Before reading the plate, ensure that there are no bubbles on the surface of the liquid.
• Substrate and stop solution should be added in the same order to eliminate any time deviations during the reaction.
