INTENDED USE 

The AFP assay is an enzyme-linked immunosorbent assay (ELISA) for the in vitro quantitative determination of alpha fetoprotein (AFP) in Human serum.

Summary

AFP is a glycoprotein with a molecular weight of between 65,000 and 70,000 Daltons including 4% of carbohydrate during fetal development, AFP maintains high levels in the serum and drops to very low levels throughout the remainder of life.AFP is elevated in the malignant diseases of hepatocelluar, testicular nonseminomatous origin, and occasionally of other endodermis origin AFP may be slightly elevated or persisted in the patients with large hepatic metastases or viral hepatitis. AFP measurement is widely accepted as tumor marker and for monitoring .the therapeutic effectiveness of hepatocellular cancer and nonseminomatous testicular cancer. The highest AFP concentration appears in the amniotic fluid in the earliest stages of pregnancy, and diminishes in concentration through the remainder of the pregnancy. Elevated AFP levels in the amniotic fluid were found in fetus with open neural tube defects, e. g. spinal bifida or anencephaly also were found in fetus with severe fetal hemolytic disease, omphalocele, esophageal atresia, congenital nephrosis, intrauterine death, or fetal bleeding into the amniotic fluid. For diagnostic purpose, ultrasonography and acetyicholinesterase measurement should be performed in conjunction with the measurement of AFP. 1,2,3 The AFP assay provides an enzyme immunoassay system with high sensitivity for human AFP measurement in serum.

Total duration of assay: 50 minutes.

The AFP assay is a quantitative solid phase enzyme-linked immunosorbent assay (ELISA).The wells are coated with anti-AFP antibodies. The samples, Standards and Controls are incubated in the wells with enzyme conjugate which is another antibody directed toward a different region of AFP molecules and chemically conjugated with horseradish peroxidase. Unbound enzyme conjugate is washed off and the amount of bound peroxidase is proportional to the concentration of the AFP present in the Samples, Standards and Controls, the intensity of color developed is proportional to the concentration of AFP in the serum, The optical density of the colored samples is read with a microplate reader at 450 nm.
 
 
Technical Parameters:

Specimen collection and preparation

• Human serum is recommended for this assay.
• Cap and store the samples at 18-25 ℃ for no more than 8 hours. Stable for 7 days at 2-8℃, and 1 month at -20℃. Freeze only once.
• Do not use heat-inactivated samples.
• Sediments and suspended solids in samples may interfere with the test result which should be removed by centrifugation. Ensure that complete clot formation in serum samples has taken place prior to centrifugation.
• Avoid grossly hemolytic, lipemic or turbid samples.
